The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. We are developing quantitative protocols and methods for characterizing polymorphisms in vertebrate genomes. We have focused on the genome of the wood frog (Rana sylvatica), the first and only completely sequenced metazoan genome. We have established techniques for assembling genomes from short-read sequence data, as well as reconstructing haplotypes from sequence data that are consistent with the sequencing data. We are using the assembled genome for our comparative analysis of genomic variation and evolution in this species. We are also investigating the pattern of genomic variation in the red-backed salamander (Plethodon cinereus), which we have assembled to 2.4x contig coverage.(1) Field of the Invention
